简介:
根据温度和压力的不同,纯净的物质会呈现出液体、气体、固体等状态的变化,在达到特定的温度和压力时,物质会出现液体与气体界面消失的现象,该点被称之为临界点。因为临界点存在的状态具有连续性,介质在液态和气态之间无明显区别,分界面的表面应力降到零,这种特性可以防止干燥样品时由于水界面急剧移动而使样品发生细微变形,避免了表面应力对试样的损坏。

临界点干燥工艺介绍:

  1. 干燥原理

根据温度和压力的不同,纯净的物质会呈现出液体、气体、固体等状态的变化,在达到特定的温度和压力时,物质会出现液体与气体界面消失的现象,该点被称之为临界点。因为临界点存在的状态具有连续性,介质在液态和气态之间无明显区别,分界面的表面应力降到零,这种特性可以防止干燥样品时由于水界面急剧移动而使样品发生细微变形,避免了表面应力对试样的损坏。利用介质临界状态(常用介质为CO2,其临界点为31.1度、72.8个大气压)下这种特性除去样品中液体的方法称之为临界点干燥法。

  1. 干燥优势

临界点干燥仪是扫面电镜生物标本制作所必须的附属设备。与烘干干燥、冷冻干燥、真空干燥等传统方法比,临界点干燥法具有被认为是目前生物扫描电镜样品制备中最可靠、最理想的干燥方法。该方法能消除液体表面张力作用,干燥处的样品最大程度的保存其自然形态,能适合多种生物样品,尤其对动物样品、含水量高且较柔的样品干燥,所以该仪器被大量用于农业微生物、畜牧、医学领域的病理疾病诊断等研究;在生物学方面用于细胞结构,以及在微型机电系统方面的样品干燥。

Step one: Sample Preparation

Sample Vitrification

This is the step which prepares the sample for cryo-TEM imaging. Vitrification cools the sample so rapidly that water molecules do not have time to crystallize, forming instead an amorphous solid that does little or no damage to the sample structure. Vitrification can be applied to protein solutions, cell suspensions or thin tissue slices.

Step two: Single Particle Imaging

Fixed in an ultra-thin vitrified ice layer, most biological materials are highly beam sensitive. As a result, only low-dose beams can be used in imaging. In the past, this has presented a challenge to acquiring sufficient image resolution and contrast. This is no longer the case.
